  • 1: DOI: 10.3892/mmr.2020.11028 2: # Prognostic value of HOXA11 expression in human cancers 3: Authors: Jiaxin Qin, Xue Miao, Gang Liang, Yinbo Chen, Ying Gao, J Chen, W Zhao, Y Gan, J Hu, B Liang, et al. 4: Journal: Molecular Medicine Reports 5: Date: May 2020 6: Keywords: homeobox A11, expression profile, survival, prognosis 7: ## Abstract 8: Dysregulated expression of HOXA11 has been involved in the development of numerous types of cancer. However, to the best of our knowledge, no studies have systematically analyzed HOXA11 expression levels in various types of cancer. The aim of the present study was to determine whether HOXA11 expression levels was associated with clinical outcomes concerning patients with different types of cancer. Firstly, the present study assessed HOXA11 mRNA expression levels by analyzing data downloaded from The Cancer Genome Atlas (TCGA) database. Next, Kaplan-Meier survival analysis was performed across all cancer types included in the TCGA database to identify whether HOXA11 expression levels were associated clinical outcome. Additionally, HOXA11 expression levels were compared between cancerous tissue and normal tissue using data from Oncomine database. Following this analysis, human protein atlas (HPA) database was queried to assess possible associations between HOXA11 protein expression levels between normal tissue and cancerous tissue. It was identified that significantly elevated HOXA11 mRNA expression levels was associated with unfavorable overall survival rates in patients with KICH (P=4×10−6), KIRC (P=9×10−12), KSARC (P=1×10−4), LIHC (P<1×10−6), LUAD (P=6×10−4), LUSC (P=4×10−4), PAAD (P<1×10−6), READ (P<1×10−6) and THCA (P=5×10−4). However, significantly reduced HOXA11 mRNA levels was associated with unfavorable overall survival rates in patients with LAML (P=7×10−4). Furthermore, increased HOXA11 mRNA expression was identified in esophageal cancer (6/7; P˂0.05), gastric cancer (8/13; P˂0.05), glioma (7/10; P˂0.05), head and neck cancer (8/8; P˂0.05), kidney cancer (12/14; P˂0.05), liver cancer (4/4; P˂0.05), lung cancer (32/42; P˂0.05), ovarian cancer (6/9; P˂0.05) and prostate cancer (9/15; P˂0.05). The present study also identified elevated HOXA11 protein expression in renal clear cell carcinoma (P˂0.05) via analysis of HPA. In conclusion, HOXA11 expression may serve a potential diagnostic biomarker of various human cancers. 9: ## Introduction 10: It is widely recognized that increasing incidence rates associated with numerous types of cancer are leading to major burdens worldwide ([1]). In recent years, advances in diagnostic strategies have led to increased use of imaging modalities and molecular markers ([2], [3]). Therefore improved identification of early-stage disease may aid efforts concerning early diagnosis strategies that may prolong survival rates. 11: The HOX family genes encode transcription factors and consist of several clusters distributed widely within human chromosome ([4]). They are critical for normal embryogenesis by determination of cell fate through controlling cell proliferation ([5]). During embryogenesis, normal HOX gene clusters are expressed during developmental stages within specific tissue domains ([6]). However, it has been consistently demonstrated that deregulated expression of HOX genes is associated with numerous types of cancer ([7]– [9]). 12: HOXA11 is located in chromosome band 7q32 ([10]), encodes transcription factors and is an important transcriptional regulator ([11]). It has been demonstrated that HOXA11 function in regulating cell cycle progression as well as cell differentiation is critical for normal embryonic development ([12]). Many studies have demonstrated that dysregulated expression of HOXA11 is involved in initiation/maintenance of various types of cancer ([13]– [15]). 13: Previous studies have reported that significantly elevated HOX-A11 mRNA levels has been described in breast cancer tissue compared with normal breast tissue ([16]– [18]). Additionally, significantly higher protein expression levels have been observed in breast cancer cell lines compared with normal breast epithelial cell lines ([17], [19]). Several studies have further demonstrated that HOXA11 protein levels were significantly correlated with increased risk of recurrence concerning patients with breast cancer ([20], [21]). Furthermore, it
